Abstract

Nanozymes have fascinated increasing attention in the field of enzyme based detection methods. In this study, we have focused on developing a rapid, colorimetric sensing method using peroxidase mimicking nanostructures for the visual detection of glucose molecules in biological samples. For this, using Fe3O4 and silk fibroin proteins, magnetic silk nanostructures (MSF nanostructures) were specifically tailored. Given their ability of catalyzing the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) in the presence of hydrogen peroxide, they can be used as colorimetric catalysts in the detection of glucose based on the production of H2O2. Using MSF nanostructures, glucose molecules were detected by both the naked eye and UV-Vis spectrophotometry in concentrations ranging from 1.4 to 86.0 μM, with a detection limit of 0.5 μM and a good linear relationship (R2=0.988). In conclusion, it can be stated that the proposed nanostructure holds great potential for biosensing applications. © 2020 Wiley-VCH Verlag GmbH & Co. KGaA, Weinheim
